Synthetic Defined (SD) / Minimal Medium This medium is considered to contain only what is essential for yeast growth. A yeast strain is considered auxotrophic if it can no longer grow on a defined medium that the wild type can grow on. This medium was used in all auxotrophic growth experiments. The medium was prepared using milli-Q grade water. It was prepared in screw bottles and autoclaved at 121 degreesC, and 15psi for 15 minutes. Contents: yeast nitrogen base (without amino acids), 6.7g/L Dextrose/D-glucose (2.5%) Supplemented with the auxotrophic requirements (e.g Met, Leu, Ura, His). G-418 sulphate (200ug/L, provided yeast strain is geneticin resistant)
